Cardiotoxicit of Human Recombinant Interleukin-2 in Rats A Morphological Study

چکیده
was given to rats for 2, 3, or 5 days. Heart, lung, liver, spleen, and kidney tissues were studied by light and electron microscopy and with immunoperoxidase techniques. Cardiac changes consisted of focal lymphocytic and eosinophilic infiltration, myocyte vacuolization, myofibrillar loss, and necrosis. Ultrastructural alterations included swelling of endothelial cells, with dissociation of intercellular junctions, migration of lymphocytes into the interstitium, and interstitial hemorrhage and edema. Close contact between infiltrating lymphocytes, particularly large granular lymphocytes, and cardiac myocytes was often observed in areas of tissue damage. All lesions were more severe on day 5 than on days 2 and 3. Immunoperoxidase stains demonstrated asialo GM, ganglioside antibody-positive, granular lymphocytes to be much more frequent in myocardium of IL-2-treated rats than in that of control rats. Conclusions. Although we cannot exclude the possibility of a direct toxic effect of IL-2 on myocytes, our observations suggest that the myocardial damage produced by this agent is triggered by IL-2-activated lymphocytes that exert cytolytic effects, first on endothelial cells and then on cardiac myocytes, thus producing lesions that involve both the cardiac microcirculation and the muscle cells. Cloning and Expression of Recombinant Human Interleukin-7 in Chinese Hamster Ovary (CHO) Cells
Background: The critical role of interleukin-7 (IL-7) in homeostatic proliferation and T cell survival has made it a promising cytokine for the treatment of various clinical conditions, especially those associated with lymphopenia. Methods: In the present study we expressed recombinant human interleukin-7 (rhIL-7) in Chinese hamster ovary (CHO)-K1 cells. CHO-K1 cells were stably transfected ...
متن کاملSimvastatin and Recombinant Antagonist of Receptors of Interleukin-1 Modulate Toll-like Receptors in Experimental Acute Ileitis in Rat
Background: The pathogenesis of inflammatory bowel disease is complex and multifactorial. Studies have led to the current concept that Toll-like receptors represent key mediators of innate host defense in the intestine, and they are involved in maintaining mucosal as well as commensal homeostasis. We studied the possibility of simvastatin and antagonist of receptors of interleukin-1 for pharmac...
متن کاملThe Effect of Mg2+ and Mn2+ on Over-Production of Interleukin-2 in Recombinant E.coli
In order to increase the productivity of human interleukin-2 (IL-2) as a model protein in the recombinant E.coliBL21 (DE3), the effect of magnesium acetate and manganese acetate was studied based on full factorial design experiments magnesium acetate and manganese acetate and their interaction exhibited an increasing effect on IL-2 expression level upto 25.24%, 16.80% and 23.30%, respective...
متن کاملBacterial Overexpression of the Human Interleukin-2 in Insoluble Form via the pET Trx Fusion System
Selection of a system for successful recombinant protein production is important. The aim of this study wasto produce high levels of human interleukin-2 (hIL-2) in soluble form. To this end, the pET32a vector inEscherichia coli BL21 (DE3) was used as an expression system, since it was previously used for the productionof mouse IL-2 in soluble form. The results indicated that c...

Background: Erythropoietin (EPO) was first known as a factor for red blood cell proliferation and differentiation. New studies show the effects of EPO on immune system. Objective: In this study, the effects of pretreatment with recombinant human erythropoietin (rHuEPO) on the anti-human leukocyte antibody (anti-HLA) titer were determined. Methods: Three groups of rats were sensitized with human...
